ABSTRACT

Objective:To investigate the role and regulatory mechanism of UGT2A3 differential expression in colorectal carcinogenesis.Methods:Nine CRC datasets were downloaded from GEO database and TCGA. R language was used to analyze the differential expression of UGT2A3 in cancer and normal tissues. According to the expression level of UGT2A3 in TCGA, the top 20 samples with the highest expression and the lowest expression were selected from normal tissues and CRC tissues, respectively. The abundance of immune cells and immune enrichment score were compared, the differentially expressed genes and differentially expressed miRNAs were screened, and the pathway enrichment analysis of differentially expressed genes was performed.Results:UGT2A3 was down regulated in all 9 CRC datasets. In all sample types, compared with the UGT2A3 high expression group, the UGT2A3 low expression group had significantly higher ImmuneScore, EstimateScoreandStromalScore, and had higher abundance of immune cells (except memory B cells). In normal tissues, the differential expression of UGT2A3 mainly affects cancer-related pathways, while in tumor tissues, it mainly affects metabolic pathways. miR-194-2, miR-224 and miR-551b were differentially expressed in all groups, which were considered as potential UGT2A3 upstream regulatory genes in CRC.Conclusions:UGT2A3, miR-194-2, miR-224 and miR-551b can be used as potential biomarkers for the diagnosis of CRC.

ABSTRACT

Objective To explore the clinical application of case teaching method combined with clinical pathway in pediatrics teaching.Methods 103 students in pediatric practice from September 2016 to September 2017 were enrolled as the study object,who were divided randomly into observation group (52 cases) and control group (51 cases).Routine teaching method was adopted in the control group,while case teaching method combined with clinical pathway was adopted in the observation group.In addition,100 cases of children in need of rescue were selected for each group and the students were divided into two or three in a group to participate in the rescue.If the student could complete the rescue with the assistance of supervisor,he will be considered as playing a leading role in rescue process.Otherwise,another supervisor would join to lead the rescue,and the student would assist by the side,who will be considered as playing an auxiliary role in rescue process.The results of examination,classroom participation,homework,the rescue performance and the satisfaction of teaching were compared between two groups.Results The scores of theoretical course,case analysis and skill operation in the observation group was significantly higher than that of the control group (P<0.05).The scores of classroom attendance,classroom activity and homework quality in the observation group was significantly higher than that of the control group (P<0.05).Success rate of rescue conducted by students in the observation group was significantly higher than that in the control group(P<0.05).The teaching satisfaction of the observation group was also significantly higher than that of the control group (P<0.05).Conclusion The application of case teaching method combined with clinical pathway teaching model in pediatrics teaching is effective and worthy of popularizing,which can obviously improve teaching quality,arouse students' interest in teaching process,and improve students' professional quality.

ABSTRACT

The PBL teaching and the case teaching have begun to be applied to the course of health economics, which has strong adaptability to the two teaching methods. Two teaching methods not only have the same characteristics, such as diversified teaching, outstanding students as the main body, paying atten-tion to the theory and practice, emphasizing on the comprehensive use of knowledge, but also have different characteristics, such as teaching idea, applied range and points of focus. These similar characteristics and differences are reflected in the development of health economics course. Based on the differences and simi-larities of the two teaching methods and further integration of the characteristics of health economics, this paper puts forward the countermeasures to optimize the application of the two teaching methods in health economics course.

ABSTRACT

Objective To evaluate the activation status of T lymphocytes and to measure the expression of OX40 and OX40L mRNA in peripheral blood from patients with systemic lupus erythematosus(SLE).Methods Peripheral blood was collected from 30 patients with SLE and 15 healthy controls.The expression of CD38 and CD3 on peripheral blood T lymphocytes were quantified by using a two-color fluorescence activated cell sorter.Fluorescence-based quantitative reverse transcription PCR was performed to determine the expression levels of OX40 and OX40L mRNA in peripheral blood mononuclear cells(PBMCs).Results The coexpression levels of CD38 and CD3 were significantly increased in patients with SLE compared with healthy controls(t =3.12,P < 0.05).Increased expression levels of OX40 and OX40L mRNA were observed in patients with active SLE compared with those with inactive SLE and healthy controls(F =4.13,3.12,both P < 0.01).The level of OX40 mRNA was positively correlated with SLE disease activity index(SLEDAI)score in the patients(r =0.35,P < 0.05).Conclusion Abnormally activated T lymphocytes and upregulated expression of OX40 and OX40L mRNA may play a certain role in the initiation and progression of SLE.
